What is Nexium 40mg?
Nexium (magnesium esomeprazole) is a proton pump inhibitor that reduces the amount of acid produced in the stomach.
Nexium is used to treat symptoms of gastroesophageal reflux disease (GERD) and other conditions associated with excessive stomach acid such as Zollinger-Ellison syndrome. It is also used to promote healing of erosive esophagitis (damage to your esophagus caused by stomach acid).
Nexium may also be used to prevent stomach ulcers caused by infection with Helicobacter pylori (H. pylori) or the use of nonsteroidal anti-inflammatory drugs (NSAIDs). Nexium is not for immediate relief of heartburn symptoms.
Important information Nexium drug 40mg
Nexium may cause kidney problems. Tell your doctor if you urinate less than usual, or if you have blood in your urine.
Diarrhea may be a sign of a new infection. Call your doctor if you have watery or bloody diarrhea.
Esomeprazole may cause new symptoms of lupus. Tell your doctor if you have joint pain and a rash in your cheeks or arms that worsen under sunlight.
You may be more likely to have a fracture while taking this medication long term or more than once daily.

Esomeprazole is the S -isome of omeprazole and reduces gastric acid secretion through a specific targeting mechanism of action. It is a specific inhibitor of acid pumps in component cells. Both R - and S -isome of omeprazole have similar pharmacodynamic activity.
Mechanism of action
Esomeprazole is a weak and concentrated base and converts into an active form in the highly acidic environment of the excretory cell of the component cell, where it inhibits the H + K + -ATPase enzyme - acid pump and inhibits both basic acid and stimulate excretion.
Pharmacodynamic effect
After taking with esomeprazole 20 mg and 40 mg, the effect begins to occur within an hour. After repeated administration with 20 mg of esomeprazole once daily for five days, the mean acid production after stimulation of pentagastrin decreased by 90% when measured 6 hours and 7 hours after dosing on the fifth day.
After five days of dosing with 20 mg and 40 mg of esomeprazole, an endometrial pH above 4 was maintained for an average of 13 hours and 17 hours, corresponding to over 24 hours in symptomatic GERD patients. The proportion of patients who maintained an internal pH above 4 for at least 8, 12 and 16 hours was esomeprazole 20 mg 76%, 54% and 24%, respectively. The respective proportions of esomeprazole 40 mg are 97%, 92% and 56%.
Using AUC as an alternative parameter for plasma concentrations, the relationship between inhibition of acid secretion and exposure has been demonstrated.
Treatment of reflux esophagitis with esomeprazole 40 mg occurred in about 78% of patients after four weeks, and 93% after eight weeks.
One week treatment with 20 mg esomeprazole bid and appropriate antibiotics, resulting in successful eradication of H. pylori in about 90% of patients.
After one week of eradication, there is no need for subsequent monotherapy with antiemetics to effectively heal the ulcer and resolve symptoms in uncomplicated duodenal ulcer.
Drug interaction Nexium 40mg
Nexium 40mg has an interaction with some other medicines. Here are some medications you should avoid concurrent use with Nexium 40mg: Diazepam; Digoxin medicine; Ketoconazole medicine; Citalopram medicine; The drug Itraconazole; Atazanavir drug; Saquinavir medicine; Atazanavir drug; Rinotavir ointment; Voriconazole medicine; Clopidogrel medicine; Warfarin medicine; Rifampicin medicine.
Note, these are not all drugs that can interact with Nexium 40mg. Therefore, users should be cautious when intending to use a combination of Nexium 40mg with any medicine.
