Protein A Resin is often used as an alternative to other methods used for the separation of protein from mixtures

Protein A—a 42-kDa chemical component found in the cell wall of staphylococcus aureus—finds wide application in biotechnological industry. This is mainly attributed to features such as its ability to bind all human IgG class immunoglobulins except IgG3.

Protein A resin can also be used for immunoprecipitation of proteins, antigens, and/or protein complexes. Protein A is derived from a strain of Staphylococcus aureus and contains five regions that bind to the Fc region of IgGThe high capacity of protein A beads makes them suitable for industrial use. Protein A media is widely used in biopharmaceutical production and related industries due to the high avidity of protein A for human IgGs. ... Not all types of antibodies will bind to protein A beads.

Protein A and G are structurally very similar, but they have slightly different affinities for IgG subclasses across different species. These affinities overlap, but in general, protein A has greater affinity for rabbit, pig, dog, and cat IgG whereas protein G has greater affinity for mouse and human IgG.

Immunoaffinity chromatography uses the specific binding of an antibody-antigen to selectively purify the target protein. The procedure involves immobilizing a protein to a solid substrate (e.g. a porous bead or a membrane), which then selectively binds the target, while everything else flows through.

Protein A Chromatography relies on the specific and reversible binding of antibodies to an immobilized protein A ligand. Protein A resins are the most frequently used affinity resins in biomanufacturing. Today it is the standard technique for capturing recombinant monoclonal antibodies. The cost of Protein A resin is nearly 50% higher than traditional chromatographic media with nonproteinaceous ligands (2, 10). Resin cost for a large column (>1-m diameter) can exceed $1 million

Protein A Resin is an affinity chromatography resin with high binding capacity for IgG. Protein A Resin is suitable for purification of monoclonal antibodies, polyclonal antibodies and immunology complexes, such as IP and Co-IP. The recombinant protein A ligand is coupled to 4% highly cross-linked agarose.
